Ingredients
Active Ingredients: clofazimine
Non-Active Ingredients: Beeswax, butylated hydroxytoluene, citric acid, ethyl vanillin, gelatin, glycerin, iron oxide, lecithin, p-methoxy acetophenone, parabens, plant oils, propylene glycol.
Indications
Lamprene is used to treat leprosy, including dapsone-resistant lepromatous leprosy and lepromatous leprosy complicated by erythema nodosum leprosum.
How It Works
The exact mechanism of clofazime is unknown at this time. However, it does bind preferentially to mycobacterial DNA, thereby it inhibits DNA replication and cell growth. Clofazimine has a slow bactericidal effect on Mycobacterium leprae and is active against various other Mycobacteria.
Dosage
Tablets (50 mg, 100 mg)
Lamprene should be used in combination with one or more other antileprosy drug. It should be taken with meals.
For dapsone-resistant leprosy: Lamprene should be given at a dosage of 100 mg daily in combination with one or more other antileprosy drugs for 2 years, followed by monotherapy with 100 mg of Lamprene daily.
Lamprene administered at dosages of 100 to 200 mg daily for up to 3 months may be useful in eliminating or reducing corticosteroid requirements. Dosages above 200 mg daily are not recommended.
The treatment of erythema nodosum leprosum will depend on the conditions.

Lamprene is excreted in mother’s breast milk, do not use medication until you consult a doctor if you are pregnant or if you are considering becoming pregnant. The safety and effectiveness in pediatric patients has not been tested, and it is best to consult a doctor before selecting any course of treatment. Physicians should be aware that skin discoloration due to Lamprene may be a sign of liver damage or failure, in which case discontinuing the medication may be suggested. Skin discoloration is also indicative of depression being caused by the medication.
Drug Interactions

Adverse/Side Effects

Common side effects include: loss of appetite, diarrhea, nausea, vomiting, dry skin and discoloration (from pink to brownish-black) of the skin, stools, urine, saliva, sweat, tears or lining of the eyelids. Other side effects reported are changes in taste, dry or irritated eyes, headaches, fever, increased blood sugar and an increased sensitivity to sunlight.
