Chemical Name
Indinavir

Ingredients
Active Ingredients:  Indinavir

Non-Active Ingredients: Anhydrous lactose and magnesium stearate. The capsule shell has the following inactive ingredients and dyes: gelatin, titanium dioxide, silicon dioxide, and sodium lauryl sulfate.

Indications
Crixivan is used in combination with antiretroviral agents is indicated for the treatment of HIV infection.

How It Works
HIV-1 protease is a contributing factor in infectious HIV-1. Indinavar is a medication that works by inhibiting the activity of enzymes the harmful enzymes. This process prevents the cleavage of viral polyproteins, which result in the creation of new, non-infectious viral particles.

Dosage
Capsules (400 mg)
The recommended initial dose of Crixivan must be taken at intervals of eight hours. Crixivan should be given without food. Water should be taken with this medication. Take Crixivan one hour before or two hours after a meal to ensure optimal absorption.

Concomitant Therapy:
Delavirdine

Dose reduction of Crixivan to 600 mg every 8 hours should be considered when administering delavirdine 400 mg three times a day.

Didanosine: should be administed one hour apart on an empty stomach.

Itraconazole: Dose reduction of Crixivan to 600 mg every 8 hours is recommended when administering itraconazole 200 mg twice daily concurrently.
Ketoconazole: dose reduction of Crixivan to 600 mg every 8 hours

Rifabutin: dose reduction of rifabutin to half the standard dose

Precautions

Urinary tact and kidney stone has happened while taking this medication. Speak to your doctor about the possibility of this happening before taking this medication.

Can cause anemia due to the abnormal breakdown of red blood cells. Once this is diagnosed, appropriate measures should be taken to institute a corrective treatment.

There have been reports of inflammation of the cell tissues in the liver. This is a very serious condition that may be fatal. Special precautions should be taken.

Hyperglycemia is noted in a few patients while taking this is medication. This means that diabetes might arise or it may result in the exacerbation of a current diabetic condition. In a few cases, Diabetic ketoacidosis has occurred. Special monitoring should take place to make sure blood sugar levels do not get out of hand.

In a few cases, patients have experienced an elevated level of the pigment bilirubin in the blood.

Patients with hepatic insufficiency due to cirrhosis should be given lower doses.

Patients with hemophilia: have sometime experienced spontaneous bleeding.

Optimal dosing regimen for use of indinavir in pediatric patients has not been established. Special precaution should be considered as there is limited data available for use on pediatric patients.

This medicine may cause harm to an unborn baby. Before taking Crixivan, tell your doctor if you are pregnant or plan to become pregnant during treatment. Crixivan  may pass into breast milk and may cause harm to a nursing baby. Consult your doctor before using this medication if you are pregnant or breast-feeding a baby.

You should consult a doctor before using this medication if you are allergic to indinavir or similar medicines, or any of the other ingredients of Crixivan .

Drug Interactions
Consult a doctor before taking Crixivan  if you are taking Midazolam (injectable), Advair, Flonase, Flovent, St. John's wort, Clarithromycin Itraconazole,  Ketoconazole,  Rifabutin,  Rifampin (Rifadin, Rimactane, Rifater), antidepressants such as trazodone (Desyrel), and others; calcium channel blocker such as Amlodipine,  Diltiazem, Felodipine, Nifedipine or Verapamil, cholesterol-lowering medicine such as Atorvastatin, Lipitor, Lovastatin, Mevacor, Altocor, Rosuvastatin, Crestor, or Simvastatin Zocor, engraf, Neoral, Sandimmune, sirolimus, Rapamune,, Tacrolimus, Prograf, sildenafil, heart rhythm medications, insulin or diabetes medication, other HIV AIDS medicine such as atazanavir etc, seizure medications such as carbamazepine etc.

Before using this Crixivan, tell your doctor or pharmacist about all prescription and non-prescription medications, including herbal products, minerals and vitamins you may be taking. Other medications may affect how Crixivan works or may cause other unforeseen complications.

Adverse/Side Effects
Infrequent side effects may include rash, steven-johnson syndrome, joint pain, abnormal skin sensation, chest pain, abnormal heart conditions, cardiovascular disorders, liver problems, hepatitis, liver failure, inflammation of the pancreas, yellowing of skin and eyes,, spontaneous bleeding, skin rash, hives, inflammation of blood cells, acute kidney failure, darkening of skin and nails, kidney infection caused by bacteria, inflammation around the nail, presence of white blood cells in urine,, bacteria or fungal infection around the hand. If these side effects persist or become overly bothersome, contact your doctor or health-care professional.

If any of these side effects become bothersome, seek medical attention immediately.
A very serious allergic reaction to this drug is rare. Get emergency medical help if you have any of these signs of an allergic reaction: rash, itching/swelling (especially of the face/tongue/throat), severe dizziness, or trouble breathing. This may not be a complete list of side effects and may be subject to change, consult with your doctor or pharmacist if you are experiencing any worrisome side effects.

Overdose
If you suspect overdose, contact your local poison control center or emergency room immediately. Symptoms of overdose include nephrolithiasis/urolithiasis, flank pain, hematuria) and gastrointestinal nausea, vomiting, diarrhea.

Storage Instructions
Store in a tightly-closed container at room temperature, 15-30°C (59-86°F) away from moisture and light, and out of reach of children and pets.
